Purification and characterization of an enzyme that degrades polyamide 4 into gamma-aminobutyric acid oligomers from Pseudoxanthomonas sp. TN-N1

Polyamide4 (PA4) is a bioplastic with superior properties and high biodegradability. Although some PA4-degrading microorganisms have been isolated from various environments, enzymes that play role in PA4 degradation are yet to be identified. In this study, we novel microorganism, Pseudoxanthomonas sp. TN-N1. This strain showed clear zone surrounding the colony on an agar plate medium containing powder. The activity using supernatant of culture was maximum when cultured appropriate 0.5% (w/v) at pH 7.0, 30 °C for 3 days. Furthermore, purified enzyme hydrolyzes amide bonds into ?-aminobutyric acid oligomers (2–4 mers) anion exchange gel permeation column chromatography. However, exhibited no protease casein, it consisted monomers or dimers molecular mass 90 kDa. optimum 7.5 35 °C, more than 80% retained range 6.0– 7.0 temperature below °C. addition, significantly inhibited by ethylenediaminetetraacetic treatment. our opinion, first report exhibiting activity. Our findings will contribute better understanding mechanism microorganisms.
